Holcomb is located at {{coord|36|24|05|N|90|01|29|W|type:city}} (36.4013424, -90.0248383).<ref name=gnis/> The city lies at the intersection of [[Missouri Route 25]] and [[Missouri Route 53]] in the western [[Missouri Bootheel|Bootheel]]. The [[St. Francis River]] and the Missouri-[[Arkansas]] state line pass just to the west.

The racial makeup of the city was 95.59% [[White (U.S. Census)|White]], 1.73% [[Black (U.S. Census)|Black]] or [[African American (U.S. Census)|African American]], 2.20% from [[Race (U.S. Census)|other races]], and 0.47% from two or more races.
