The region was first settled ''circa'' 1782. The town of Barker was formed in 1831 from a portion of the town of [[Lisle, New York|Lisle]].

The town is bordered to the north by the town of [[Triangle, New York|Triangle]], to the northwest by [[Lisle, New York|Lisle]], the west by [[Nanticoke, New York|Nanticoke]], the southwest by [[Maine, New York|Maine]], the south by [[Chenango, New York|Chenango]], and the southeast by [[Fenton, New York|Fenton]]. The eastern town line is the border of [[Chenango County, New York|Chenango County]] (town of [[Greene, New York|Greene]]).

[[Interstate 81 in New York|Interstate 81]], [[U.S. Route 11 in New York|U.S. Route 11]], [[New York State Route 79]], and the [[Tioughnioga River]] pass through Barker. The population density was {{convert|66.2|PD/sqmi|PD/km2|sp=us|adj=off}}.  There were 1,067 housing units at an average density of {{convert|25.8|/sqmi|/km2|sp=us|adj=off}}.  The racial makeup of the town was 97.77% [[White (U.S. Census)|White]], 0.77% [[African American (U.S. Census)|African American]], 0.11% [[Native American (U.S. Census)|Native American]], 0.33% [[Asian (U.S. Census)|Asian]], 0.15% from [[Race (United States Census)|other races]], and 0.88% from two or more races. The average household size was 2.76 and the average family size was 3.10.
