Major roads include two U.S. Highways: [[U.S. Route 80 in Alabama|U.S. Route 80]] (US 80) runs east–west through the city and [[U.S. Route 43|US 43]] runs north–south. Demopolis is served by several railway companies, including [[Norfolk Southern Railway]], [[BNSF Railway]], and the [[Alabama and Gulf Coast Railway]].

The [[Demopolis Municipal Airport]] is located northwest of the city, adjacent to Airport Industrial Park and the Tennessee-Tombigbee Waterway.
