Ladson is located in southwestern Berkeley County, northern Charleston County, and southeastern Dorchester County at {{coord|33|0|34|N|80|6|20|W|type:city}} (33.009563, -80.105553).<ref name="GR1">{{cite web|url=https://www.census.gov/geographies/reference-files/time-series/geo/gazetteer-files.html|publisher=[[United States Census Bureau]]|access-date=2011-04-23|date=2011-02-12|title=US Gazetteer files: 2010, 2000, and 1990}}</ref> It is bordered to the southwest by the town of Summerville, to the south by the city of North Charleston, to the east by the city of [[Goose Creek, South Carolina|Goose Creek]], and to the northwest by [[Sangaree, South Carolina|Sangaree]], a census-designated place.

[[U.S. Route 78]] and [[Interstate 26]] run parallel through Ladson, with Exit 203 providing access from I-26. Downtown [[Charleston, South Carolina|Charleston]] is {{convert|20|mi}} to the southeast, and [[Columbia, South Carolina|Columbia]] is {{convert|97|mi}} to the northwest.

According to the [[United States Census Bureau]], the CDP has a total area of {{convert|18.2|sqkm|order=flip}}, all land. This is a decrease from {{convert|22.3|sqkm|order=flip}} at the 2000 census, due to annexations of portions of the area into [[Summerville, South Carolina|Summerville]] and [[North Charleston, South Carolina|North Charleston]].
